Scaffolds are usually rigid, hetero-cycle based structures
with functional groups that can be further modified. A large
number of marketed drugs contain core scaffolds, for example,
Atorvastatin, Sildenafil, and Erlotinib, to name a few. It
appears that the major role of the scaffold in a drug molecule
is to position those attached functional groups in a right
direction and facilitate their proper interaction with biological
targets. Therefore, tuning scaffolds is a proven strategy to
discover drugs with improved therapeutic index.
